Widespread use of TATA elements in the core promoters for RNA polymerases III, II, and I in fission yeast.
In addition to directing transcription initiation, core promoters integrate input from distal regulatory elements. Except for rare exceptions, it has been generally found that eukaryotic tRNA and rRNA genes do not contain TATA promoter elements and instead use protein-protein interactions to bring the TATA-binding protein (TBP), to the core promoter. Genomewide analysis revealed TATA elements i...
متن کاملSynthetic Core Promoters for Pichia pastoris
Synthetic promoters are commonly used tools for circuit design or high level protein production. Promoter engineering efforts in yeasts, such as Saccharomyces cerevisiae and Pichia pastoris have mostly been focused on altering upstream regulatory sequences such as transcription factor binding sites. In higher eukaryotes synthetic core promoters, directly needed for transcription initiation by R...
متن کاملA discrimination study of human core-promoters.
A core-promoter, approximately from -60 bp upstream to +40 bp downstream of a RNA polymerase (RNAP) II transcription start site (TSS), binds to the preinitiation complex (PIC) and determine the position of TSS. Using position-specific k-tuple feature variables, a quadratic discriminant analysis (QDA) method is shown to be very effective in identifying human core-promoters.
متن کاملViral Enhancer Mimicry of Host Innate-Immune Promoters
The inflammatory milieu is the natural habitat for a pathogenic infection, characterised by activity of pro-inflammatory signalling pathways and inflammatory cytokines. Viral entry rapidly activates a range of innate-immune signalling events such as the activation of Pattern Recognition Receptors (PRRs) [1–5]. A virus must therefore counteract intrinsic cellular and innate-immune responses to s...
متن کاملExome-based Variant Detection in Core Promoters
Core promoter controls the initiation of transcription. Core promoter sequence change can disrupt transcriptional regulation, lead to impairment of gene expression and ultimately diseases. Therefore, comprehensive characterization of core promoters is essential to understand normal and abnormal gene expression in biomedical studies. Here we report the development of EVDC (Exome-based Variant De...
